 B1-driven rapid non-transcriptional responses. This study reveals function
 al specialization between homologous receptors: TIR1 operates in the nucle
 us for transcriptional activation\, while AFB1 mediates cytoplasmic rapid 
 responses via subcellular compartmentalization and intrinsic biochemical f
 eatures. Nuclear-localized AFB1 fails transcriptional regulation due to de
 fective SCF complex assembly.Crucially\, TIR1’s adenylate cyclase activi
 ty generates cAMP as a secondary messenger. cAMP activates ARFs independen
 tly of Aux/IAA degradation by directly binding ARFs\, promoting their diss
 ociation from repressors and enhancing homodimerization. Genetic and bioch
 emical analyses confirm cAMP-ARF interaction is essential for transcriptio
 nal activation.These findings establish a parallel TIR1-cAMP-ARF axis\, re
 vising the classical auxin signaling model. The study uncovers receptor sp
 ecialization through spatial segregation and biochemical divergence\, whil
 e identifying cAMP as a novel mediator linking auxin perception to transcr
 iptional reprogramming. This dual regulatory framework advances understand
 ing of hormonal signaling complexity in plants.
