DESCRIPTION:Speaker: Maria Leptin\nAbstract: The terminal cells of the Dros
 ophila respiratory (tracheal) network contain seamless\,\nmembrane bounded
  intracellular tubes through which air travels. The tube-containing\nexten
 sions of these cells are elaborated during chemotactic growth directed by\
 ndevelopmental cues in embryos and subsequently ramify in response to sign
 als from\nhypoxic tissues in larvae. Extensive membrane traffic occurs dur
 ing growth\, as the cell\nrapidly and concomitantly elaborates two membran
 e domains of opposite characteristics.\nThe outer (basal) membrane migrate
 s and grows with highly dynamic filopodial extensions\,\nwhile at the same
  time the inner tube of apical characteristics is constructed from as yet\
 nunknown membrane sources.\nWe find that the membranes forming the intrace
 llular tubules contain lipids and proteins\ntypical of apical plasma membr
 anes in polarized epithelial cells. The Drosophila\nsynaptotagmin-like pro
 tein Bitesize (Btsz) and the activated form of its interaction partner\nMo
 esin are also located at the growing luminal membrane. Our functional stud
 ies indicate\nthat the actin cytoskeleton\, through its interaction with B
 tsz via Moesin directs apical\nmembrane morphogenesis to create and mainta
 in distinct intracellular tubules.\nUsing real-time in vivo imaging we hav
 e analysed the assembly of the intracellular tube.\nWe find that ER and Go
 lgi rapidly distribute into the developing cellular extensions prior to\nt
 he assembly of the membrane bounded tube within. Redistribution of secreto
 ry\nmachinery ahead of the growing tube seems to be a prerequisite for pro
 per tube\nextension.
