DESCRIPTION:Speaker: Thomas Silhavy\nAbstract: The outer membrane (OM) of G
ram-negative bacteria functions as a protective barrier. It\nis unusual be
cause the OM bilayer is asymmetric\; the inner leaflet is composed of\npho
spholipids\, but the outer leaflet is made of lipopolysaccharide (LPS). Tw
o kinds of\nproteins are found in the OM. Lipoproteins are inserted into t
he inner leaflet of the OM by\nposttranslationally attached lipid moieties
. Integral OM proteins are b-barrel proteins\n(OMPs). We have used genetic
\, bioinformatic and biochemical strategies to identify and\ncharacterize
the cellular components required for LPS and OMP biogenesis. After\ndescri
bing what is known about these essential cellular processes\, I will focus
on the\ngenetic approaches we are taking to probe the mechanism by which
?-barrel assembly\nmachine folds and inserts OMPs into the OM. This machin
 e is composed of a large ?-barrel\nprotein\, BamA\, and four lipoproteins\
, Bam BCDE. My talk will focus on the role of BamE\nand how the two essent
ial proteins\, BamA and BamD communicate with each other.
